Uncovering cell of origin dependent mechanisms of transformation in Kras mutant malignancies

Our lab has developed acinar and ductal derived mouse models of ductal adenocarcinoma.
Employing scRNAseq, RNAseq, proteomic analysis and whole exome sequencing, our lab is studying mechanistic differences in acinar versus ductal cell transformation.
PanIN and Stromal Heterogeneity
Our lab has published mouse models of ductal and acinar derived PanIN. Using RNAseq on ex vivo cultured ductal and acinar cells, our lab has discovered early mechanistic drivers of Kras mutant PanIN. Our lab is now studying cell autonomous and paracrine drivers of PanIN and stroma using genetically engineered mouse models.
Metabolic reprogramming in lung adenomas and non-small cell lung cancer
Using Proteomic Analysis and transgenic mouse models, we recently published Hippo signaling regulates expression of PKM2 (Singh et al., 2020).
Using metabolic profiling and RNAseq, we are expanding on mechanistic data described in this publication to understand metabolic reprogramming in Kras mutant non small cell lung cancer.
